ABSTRACT

@#[摘要] 目的:寻找多发性骨髓瘤(MM)基因芯片数据集中的潜在致病基因、疾病诊断和预后相关因子并阐明其作用机制。 方法:获取基因芯片数据集GSE13591 和Zhan Myeloma,使用R 语言进行基因差异表达分析。对共同高表达基因进行Meta 分 析,得到P 值中位秩次TOP10 基因。CCLE 数据库分析TOP10 基因在各肿瘤细胞系中的mRNA表达情况,筛选出蛋白酶体通路 相关基因8(DCAF8)。cBioPortal 数据库中分析DCAF8 基因在各肿瘤细胞系的突变率。WB检测DCAF8 蛋白在骨髓瘤细胞系中 的表达情况。SPSS 和Graph Pad 软件分析DCAF8 表达量和MM患者临床特征之间的相关性,进行受试者工作特征曲线(ROC曲 线)绘制和生存分析。R语言Custer Profiler Package 和Metascape 数据库对DCAF8 互作蛋白基因和共表达基因进行GO和KEGG 功能富集分析。结果:数据集GSE13591 和Zhan Myeloma 的上调基因取交集得到477 个共同高表达基因。在合并数据集中对上 述基因进行Meta 分析,得到P 值中位秩次TOP10 基因,DCAF8 在MM细胞系中的平均表达水平最高。DCAF8 基因在浆细胞骨 髓瘤中的突变率位于各肿瘤细胞系的第一位。DCAF8 蛋白在多种MM细胞系中的表达量显著升高(P<0.01)。DCAF8 表达量与 MM患者的肿瘤负荷显著正相关,1q21 扩增阳性组的DCAF8 的表达量显著高于1q21 阴性组。ROC曲线显示DCAF8 的表达水平 可以很好地区分MM患者和正常人(P<0.01)。DCAF8 高表达组比DCAF8 低表达组中位生存时间显著缩短。蛋白互作网络显示 DCAF8 可与XPO1 蛋白直接相互作用。GO和KEGG功能富集分析结果表明,DCAF8 与蛋白酶体功能、剪切体活性、组蛋白乙酰 化酶活性、RNA转运等功能相关。结论:DCAF8 在MM中显著高表达,其表达水平可以很好地区分MM患者和正常人;其高表 达与MM患者的生存预后显著负相关,且与1q21 扩增和XPO1 蛋白相关。

ABSTRACT

In the present study, we aimed to compare the detoxifying effects of two fat emulsions containing either long-chain triglyceride or a mixture of medium-chain and long-chain triglycerides in the propafenone-poisoned rat model. Rats were randomly divided into 3 groups according to the fat emulsions used: long-chain triglyceride-based fat emulsion (LL) group; medium-chain and long-chain triglyceride-based fat emulsion (ML) group; normal saline (NS) group. Propafenone was continuously pumped (velocity=70 mg/kg per h) until the mean blood pressure dropped to 50% of basal level. Then, LL/ML fat emulsions or NS was intravenously infused instantly with a loading-dose (1.5 mL/kg) and a maintenance dose (0.25 mL/kg per min) for 1 h. Subsequently, the propafenone was added to plasma (3.5 μg/mL) in vitro, mixed with three doses of LL or ML (1, 2, or 4%). Finally, after centrifugation, the concentration of propafenone was measured. Rats treated with LL exhibited accelerated recovery, characterized by higher blood pressure and heart rate. Rats in both the LL and ML groups demonstrated decreased propafenone in plasma (time-points: 15, 25, and 60 min). However, rats that received LL showed lower propafenone in myocardial tissue at the end of detoxification treatment. Rats in the ML group had the lowest value of pH, the minimum content of HCO3-, and the highest production of lactic acid at the end. In the in vitro experiments, propafenone decreased more dramatically in the LL group compared to the ML group. Long-chain triglyceride fat emulsion had a better effect on treating propafenone poisoning in rats.

ABSTRACT

AIM:To analyze of refractive status after vitrectomy combined with phacoemulsification in patients with vitreoretinal disease and cataract.METHODS:A total of 150 patients with vitreoretinal disease and cataract were treated in our hospital from January 2014 to November 2016.According to the random number table method they were divided into two groups:combination group (75 cases) with vitrectomy combined with phacoemulsification,the control group (75 cases)with vitrectomy first,then phacoemulsification surgery.The recovery of visual acuity and the change of the axial length of the eyes were observed.The patients were followed up for 6mo,and the postoperative complications were recorded.RESULTS:After treatment,number of patients in the two groups with visual acuity ≥ 0.1 were significantly improved compared with before treatment (P<0.05),and the percent in combination group was 95%,higher than 63% in the control group (P<0.05).There were no significant changes in the axial length before and after the operation in the two groups (P>0.05),and there was no significant difference between the groups before and after treatment (P>0.05).The refractive status of combination group shifted to myopia,that of control group shifted to hyperopia,two groups had no statistically significant difference on numerical prediction (P < 0.05).The difference was statistically significant on the actual values (P<0.05).The complication rate in the combination group was 24%,which was lower than 40% in the control group,and the difference was statistically significant (P<0.05).CONCLUSION:Vitrectomy combined with phacoemulsification on vitreoretinal diseases with cataract patients is effective,and safety,clinical application value is higher.

ABSTRACT

Human contrast sensitivity is considered in relation to the responses delivered by retinal ganglion cells of the primate to luminance and chromatic contrast. At different temporal frequencies, response amplitude relative to response variability determines the limit to sensitivity of a single ganglion cell. This can be related to specific models of central detection mechanisms. Both for luminance and chromatic contrast, psychophysical sensitivity to temporal modulation can be achieved by summation of activity of just a few cells that provide input to a detection mechanism. This analysis is then extended to a spatial context. Several sets of data indicate that, in spatial terms, detection mechanisms are of limited spatial extent, and that, at least in the case of luminance patterns, eye movements play a critical role in contrast detection.
